    1. My motivation for researching this topic comes from the direct impact our work can have on the world, especially during a global pandemic. The benefits of developing a more sustainable alternative to vaccine cold chain include minimizing waste, reducing costs, and increasing access to vaccines.

  1. What effect do LDAO and tween 20 have on the secondary/tertiary structure on the protein in the vaccine? Does it retain viability as a vaccine after being desorbed from the silica gel?

    1. Great question! The CD analysis in Figure 5 indicates a small decrease in molar ellipticity for IpaD heated on silica compared to its native CD spectra when using LDAO as the desorbing agent. This small decrease should not reduce IpaD’s efficacy in a vaccine, but we are continuing to evaluate other mild detergents (e.g. Tween 20) in order to maximize protein stabilization.

  3. Love your CD curves looking at secondary structure. Would it be possible to do a temperature based CD curve, where the secondary structure is measured as the temperature increases over time? This may help show how the secondary structure is lost as the temperature increases
